Uncharacterized Protein Targets refers to a broad category of proteins identified through genomic sequencing and proteomic analysis that lack experimental annotation regarding their biological function, biochemical activity, or role in disease (UniProt, 2023). These proteins constitute a significant portion of the human proteome, often termed the 'dark proteome,' and represent a major frontier in drug discovery (Nature Reviews Drug Discovery, 2018). While individual uncharacterized proteins may eventually be validated as therapeutic targets, the term itself is a placeholder for gene products with unknown mechanisms (NIH IDG, 2024). Research initiatives like the NIH's 'Illuminating the Druggable Genome' (IDG) project aim to characterize these proteins to identify novel therapeutic opportunities (Common Fund, 2024). Currently, because their structures and ligands are largely unknown, they are generally not targeted by existing pharmacological agents. However, advances in artificial intelligence and chemoproteomics are increasingly being used to predict their functions and identify potential small-molecule binders (BioRxiv, 2024).
