Unconventional SNARE in the endoplasmic reticulum 1 (USE1) is an integral membrane Qc-SNARE protein primarily located in the endoplasmic reticulum. USE1 mediates vesicle fusion events, particularly in the retrograde transport pathway from the Golgi apparatus to the endoplasmic reticulum (ER). It forms part of a SNARE complex essential for the targeting and fusion of Golgi-derived vesicles with the ER, working in concert with other SNAREs including Ufe1, Sec20, and Sec22. In mammalian systems, USE1 is involved not only in retrograde but also post-Golgi transport, and it plays a critical role in the proper glycosylation and maturation of both cellular and certain viral (e.g., mumps virus F protein) membrane proteins. Dysfunction or knockdown of USE1 impairs complex N-linked glycosylation and affects protein expression at the cell surface, with cell-type-specific consequences. Disease associations have been reported with certain types of cancer and heart disease, but USE1 is not considered a direct therapeutic target or receptor.
