Uncoordinated-119 lipid binding chaperone (UNC119, also known as HRG4) is a lipid-binding cargo adapter protein found in many species, including humans, where it is encoded on chromosome 17[2][1][4][6]. UNC119 specifically binds to myristoylated proteins and is required for their correct localization, especially to cilia and photoreceptor synapses in the retina[1][2][3][6]. This protein plays a key role in processes such as cilium assembly, protein trafficking, and the maintenance of photoreceptor function in the eye. It is also implicated in synaptic signaling and neuronal development, immune cell signal transduction (as an activator of Src family kinases in T cells), and endosome recycling[2][6]. Mutations in UNC119 are associated with disorders such as retinitis pigmentosa and Joubert syndrome, and may affect immune function[1][2]. There are no known drugs directly targeting UNC119, nor is it currently validated as a therapeutic target, but it is a key component of intracellular lipid-binding transport systems.
