Unknown kinase

The term "Unknown kinase" is a generic placeholder used in biological research and biochemical databases to refer to a protein kinase whose specific molecular identity, sequence, or function has not yet been identified [13]. In signaling pathways, it often denotes an observed phosphorylation event where the responsible enzyme remains elusive, such as the phosphorylation of Dishevelled (DVL) in the Wnt pathway [13]. This term also relates to the "dark kinome," a subset of the human genome containing approximately 160 kinases that are poorly understood and lack specific chemical probes or well-defined physiological roles [5]. Because it does not represent a single, unique therapeutic target, it cannot be specifically modulated by drugs, and its biological significance is entirely dependent on the specific substrate or pathway context in which the activity is observed [2, 6].
