Overview

The "Unknown novel inflammation checkpoint" is a first-in-class therapeutic target identified by Nucleome Therapeutics as a master orchestrator of inflammation resolution. Discovered using proprietary 3D human genetics technology (Micro Capture-C), the target was linked to causal genetic variants in the non-coding genome that are associated with chronic inflammatory diseases. In healthy individuals, this checkpoint acts to restore immune balance by promoting the suppressive activity of regulatory T cells (Tregs) while simultaneously inhibiting the activation of B cells, T cells, and macrophages. In patients with autoimmune conditions such as rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and inflammatory bowel disease (IBD), this mechanism becomes dysregulated, leading to persistent and unresolved inflammation. Therapeutic intervention with agonist monoclonal antibodies, such as NTP464, is designed to stimulate this checkpoint to actively resolve inflammation, offering a novel principle in immunology that moves beyond traditional immunosuppression.
