Upregulated lung cancer 10 (URLC10) is a cancer/testis antigen that is highly expressed in various malignancies, particularly non-small cell lung cancer (NSCLC), small cell lung cancer (SCLC), and pancreatic cancer, while remaining virtually absent in normal adult tissues except for the testis (PubMed: 15343322). It plays a significant role in promoting cell proliferation and survival, and its knockdown has been shown to inhibit the growth of cancer cells (PubMed: 15343322). Due to its restricted expression pattern and essential role in tumor cell viability, URLC10 is considered an ideal target for cancer immunotherapy, specifically for the development of peptide-based vaccines (PubMed: 21858018). Therapeutic strategies currently focus on using URLC10-derived peptides to elicit a specific cytotoxic T-lymphocyte (CTL) response to eliminate tumor cells expressing the antigen (PubMed: 21858018). Clinical trials have explored its use in combination with other cancer/testis antigens to treat advanced cancers, demonstrating its potential as a viable therapeutic target (PubMed: 24633677).
