Overview

Upstream signaling pathways encompass the initial biochemical events in a signal transduction cascade, typically initiated by the binding of ligands to cell surface receptors such as Receptor Tyrosine Kinases (RTKs) or G Protein-Coupled Receptors (GPCRs) (Cooper, 2000). These pathways involve the activation of early-stage intracellular mediators, including small GTPases like RAS and lipid kinases like PI3K, which relay signals from the extracellular environment to downstream effectors (Nature Education, 2014). Because the term refers to a collective group of processes rather than a single protein, it is not a specific therapeutic target; however, many individual components within these upstream pathways are major focuses of drug development. Dysregulation of upstream signaling is a primary driver in various pathologies, most notably in oncology, where mutations leading to the constitutive activation of these pathways promote autonomous cell growth and survival (Hanahan & Weinberg, 2011). Targeting specific nodes within these upstream cascades is a common strategy in precision medicine to interrupt pathological signaling while minimizing broader systemic disruption.
