Overview

Upstream stimulatory factor 2 (USF2) is a ubiquitously expressed basic helix-loop-helix leucine zipper transcription factor that modulates gene expression by binding conserved E-box motifs in DNA. USF2 participates in the regulation of diverse biological processes including cell proliferation, differentiation, metabolism, stress response, immune function, and tissue-specific gene regulation. Dysregulation of USF2 contributes to enhanced proliferation and immortality in cancer cells, influences steroid biosynthesis in endocrine tissues, and is linked to pathological states such as fibrosis, hepatocellular carcinoma, and endometriosis. USF2 interacts with other transcription factors (notably USF1) and responds to both environmental and cellular cues, positioning it as a pivotal node in transcriptional control, but it is not established as a direct therapeutic target or receptor[1][2][5].
