Overview

Uridine phosphorylase (UPase, EC 2.4.2.3) is an enzyme that catalyzes the reversible phosphorolysis of uridine to uracil and alpha-D-ribose 1-phosphate. It belongs to the glycosyltransferase family and plays a central role in pyrimidine metabolism by participating in the salvage pathway for pyrimidine nucleosides. Its activity is often elevated in various tumor tissues, enhancing sensitivity to chemotherapeutic agents like 5-FU. Inhibitors of UP or genetic disruption can modulate drug toxicity profiles or rescue normal tissues from chemotherapeutic side effects.
