Overview

Uroplakin-1b (UPK1B) is a tetraspanin family integral membrane protein that is a structural component of the asymmetric unit membrane (AUM) forming plaques on the apical surface of urothelial cells in the bladder and kidney. These plaques contribute to the permeability barrier of the urothelium, maintaining structural integrity during bladder distension and protecting superficial cells from urine exposure. UPK1B plays a vital role in urothelial differentiation, tissue development, and homeostasis, and loss or dysregulation of UPK1B leads to urothelial plaque disruption, altered cell composition, dysplasia, and congenital anomalies of the urinary tract. UPK1B is not a classical drug target (such as a receptor, enzyme, or transporter), but it is relevant as a disease marker and as a structural protein essential for urinary tract biology.
