The urothelial surface refers to the apical (luminal) layer of the urothelial lining, primarily composed of large, flattened umbrella cells interconnected by tight and adherens junctions. This surface is covered by specialized protein plaques (asymmetric unit membrane), mainly made of uroplakins, and forms one of the body’s tightest permeability barriers, crucial for containing urine and protecting against pathogens and toxins. The urothelial surface is not a single molecular target, but incorporates numerous specific proteins and receptors (including uroplakins, various ion channels, and GPCRs), each potentially representing a drug target or biomarker in its own right[3][5][6][7][8]. Damage or dysfunction at the urothelial surface underlies a number of urological diseases, including bladder cancer, interstitial cystitis, and urinary tract infections.
