Overview

USP2 antisense RNA 1 (USP2-AS1) is a long non-coding RNA (lncRNA) with strong pro-oncogenic functions demonstrated in multiple cancer types, including head and neck squamous cell carcinoma, hepatocellular carcinoma, ovarian cancer, colon cancer, and lung cancer[1][4][6][7]. USP2-AS1 is transcriptionally regulated by oncogenic factors such as HIF1α (under hypoxic conditions) and c-Myc[1][3][4]. It promotes cancer cell proliferation, cell cycle progression, invasion, and inhibits cellular senescence, mainly by interacting with regulatory proteins such as DCAF13, G3BP1, and YBX1, leading to alterations in the stability and translation of key mRNAs involved in tumorigenesis (e.g., E2F1, HIF1α)[1][3][4]. High levels of USP2-AS1 are associated with poor prognosis in several cancers and it is being investigated as a biomarker for cancer prognosis and as a potential therapeutic target, though there are currently no known direct drug interactors or approved drugs targeting this lncRNA[1][4][6][7].
