Uterine smooth muscle tone refers to the contractile state of the myometrium, which is essential for reproductive processes including menstruation, pregnancy maintenance, and parturition (StatPearls, 2023). This physiological state is regulated by a variety of molecular targets, most notably the oxytocin receptor (OXTR) and prostaglandin receptors (EP and FP), which increase intracellular calcium to promote contraction (PubMed, 2019). Conversely, relaxation is mediated by beta-2 adrenergic receptors and nitric oxide pathways that increase cyclic AMP or cyclic GMP levels (NIH, 2021). Clinical management of uterine tone is vital; uterotonic drugs like oxytocin and misoprostol are used to induce labor or treat uterine atony, a major cause of postpartum hemorrhage (WHO, 2018). Tocolytic agents, such as nifedipine and atosiban, are employed to suppress preterm labor by inhibiting these contractile pathways (PubMed, 2020). Because Uterine smooth muscle tone describes a complex physiological outcome involving multiple signaling systems rather than a single molecule, it is classified as a biological process rather than a specific therapeutic target.
Modulation occurs via agonism or antagonism of G protein-coupled receptors (e.g., oxytocin and prostaglandin receptors), inhibition of calcium influx through voltage-gated channels, or activation of beta-adrenergic signaling pathways to alter intracellular calcium levels and myosin light chain kinase activity.
