Overview

V-set and immunoglobulin domain-containing protein 10-like (VSIG10L) is a single-pass membrane protein of the immunoglobulin superfamily, characterized by extracellular immunoglobulin (Ig)-like C2 domains, a transmembrane region, and a small cytoplasmic tail[3][4]. It is highly expressed in the healthy squamous epithelium of the esophagus, where it plays a critical role in epithelial maturation and maintaining tissue integrity[1][2][3]. Germline mutations such as S631G are associated with familial susceptibility to Barrett esophagus and esophageal adenocarcinoma, as they impair epithelial differentiation and cell-cell adhesion[1][2]. Loss of VSIG10L expression in esophageal lesions is a marker for pathological change, and rare somatic mutations have also been observed in cancer tissue[1]. The specific function may include immune modulation, as recombinant VSIG10L can inhibit IFN-gamma secretion by human immune cells[3]. VSIG10L belongs to the broad VSIG protein family, which is involved in cell adhesion, immune response, and signal transduction[4]. Its role as a therapeutic target is not established, and there are no approved drugs targeting VSIG10L.
