V-set and immunoglobulin domain-containing protein 2 (VSIG2) is a single-pass type I transmembrane protein characterized by extracellular immunoglobulin C2 and V-set domains and a cytoplasmic tail[1][5]. Initially identified as a marker on cortical thymocytes in non-mammalian species, VSIG2 in humans and mice is abundantly expressed in tissues such as thyroid, trachea, prostate, colon, and stomach, but shows weak or no expression in the thymus[1]. Its structure suggests a possible function in cell adhesion, similar to junctional adhesion molecules, but its specific physiological roles are not well characterized[4][2]. Evidence from cancer studies indicates that VSIG2 may function as a tumor suppressor, particularly in colorectal cancer, where low expression is associated with poor prognosis, and its abundance correlates with immune cell infiltration[1]. However, VSIG2 is not currently established as an immune checkpoint or as a validated pharmacological target[1].
