VSIG4 (V-set and immunoglobulin domain-containing protein 4), also known as CRIg, is a type I transmembrane protein predominantly expressed on tissue-resident macrophages. It functions as both an immune checkpoint molecule, negatively regulating T cell activation, and a complement receptor, binding C3b/iC3b to mediate phagocytosis and inhibit the alternative complement pathway. Its restricted expression pattern and dual functions make it a potential therapeutic target in autoimmunity, infectious disease, and cancer.
