Overview

Vacuolar protein sorting-associated protein 26A (VPS26A) is a subunit of the retromer complex, a multimeric protein complex crucial for the recycling and retrograde trafficking of membrane proteins from endosomes back to the trans-Golgi network or plasma membrane[3][4]. VPS26A forms a core cargo recognition complex (CRC) with VPS29 and VPS35, engaging in cargo selection and facilitating retromer assembly[3]. The protein has a two-lobed beta-sandwich structure with an arrestin-like fold and interacts directly with VPS35, integrating into dynamic subcomplexes within the retromer[1][3][4]. Dysfunction or mutations in retromer components, including VPS26A, have been linked to impaired protein sorting and are implicated in the pathology of some neurodegenerative diseases, highlighting its biological importance but not its direct role as a therapeutic target[3].
