Vacuolar protein sorting-associated protein 26B (VPS26B) is a peripheral membrane protein that forms part of the retromer complex, a multi-protein assembly critical for endosomal protein sorting and retrograde transport from endosomes to the trans-Golgi network or plasma membrane[1][2][4][5]. In mammals, VPS26B is one of two paralogs of VPS26 (the other is VPS26A); VPS26B is notably enriched in the central nervous system and forms a cargo-recognition trimer with VPS35 and VPS29, creating a retromer subcomplex[1][4]. VPS26 proteins show structural similarity to arrestin family members, but VPS26B lacks specific arrestin interaction sites[4][6]. VPS26B is involved in the trafficking and recycling of key neuronal proteins, such as the amyloid precursor protein (APP), implicating it in neurodegenerative disease mechanisms[1][8]. Current evidence does not indicate that VPS26B is a direct therapeutic target (such as a receptor or enzyme), and there are no known drugs directly targeting this protein.
