Overview

The VPS35–VPS29 subcomplex is part of the retromer cargo recognition core, which mediates selective retrieval and recycling of transmembrane proteins from endosomes to the trans-Golgi network or cell surface. VPS35 forms a right-handed α-helical solenoid that wraps around VPS29, whose metallophosphoesterase-like domain acts as a scaffold rather than a functional enzyme. This subcomplex is essential for proper sorting of lysosomal enzymes, maintenance of cellular homeostasis, and is implicated in neurodegenerative disorders when mutated or dysfunctional. It does not form an independent functional unit but requires interaction with VPS26 and membrane-targeting SNX proteins as part of the retromer complex
