Vacuolar protein sorting-associated protein 37A (VPS37A) is a subunit of the ESCRT-I (endosomal sorting complex required for transport I) complex[4]. This complex is crucial for sorting ubiquitinated membrane proteins into multivesicular bodies destined for lysosomal degradation, and VPS37A specifically contributes to complex assembly, cargo sorting, and membrane scission events[2][1]. VPS37A is involved in receptor down-regulation (e.g., EGFR), autophagosome closure during autophagy, and has roles in vesicular trafficking critical for processes such as viral budding (e.g., HIV)[1][2][3]. Dysregulation or mutation of VPS37A can contribute to disease, including hepatocellular carcinoma (where it is downregulated) and autosomal recessive spastic paraplegia type 53[4]. There are no therapeutic drugs or mechanisms directly targeting VPS37A currently identified.
