Vacuolar protein sorting-associated protein 37B (VPS37B) is a subunit of the ESCRT-I (Endosomal Sorting Complex Required for Transport-I) complex, which plays a central role in sorting ubiquitinated membrane proteins into multivesicular bodies for lysosomal degradation. VPS37B participates in vesicular trafficking, specifically the endosomal sorting pathway, protein transport, and assembly of multivesicular bodies. It is important for the cellular process of targeting endocytic cargo, facilitating viral budding (such as HIV-1), and is necessary for maintaining ESCRT-I complex stability. Alterations in VPS37B expression, particularly decreased expression, have been implicated in the progression of certain cancers, such as colorectal cancer, and can provoke cellular stress responses.
