Vacuolar protein sorting-associated protein 37D (VPS37D) is a structural subunit of the ESCRT-I (Endosomal Sorting Complex Required for Transport-I) complex, a key regulator of vesicular trafficking in eukaryotic cells. The ESCRT-I complex is essential for sorting ubiquitinated endocytic cargos into multivesicular bodies (MVBs), which are specialized endosomes involved in cargo sorting, membrane remodeling, and regulation of receptor and protein degradation. VPS37D participates in this process as one of several subunits that form the heterotetrameric ESCRT-I complex. In addition to its canonical role in vesicular transport, defects in VPS37D are associated with Williams-Beuren syndrome and certain developmental disorders. VPS37D is not considered a therapeutic target, receptor, enzyme, transporter, or clinically actionable drug target, and no drugs are currently known to interact with it[2][3][5].
