Overview

Vacuolar protein sorting-associated protein 52 homolog (VPS52) is a core subunit of the tetrameric **Golgi-associated retrograde protein (GARP) complex**, which mediates **retrograde vesicle tethering and fusion from endosomes to the trans-Golgi network** in eukaryotic cells. The GARP complex belongs to the CATCHR family of multisubunit tethering complexes and functions as an obligate heterotetramer with VPS51, VPS53, and VPS54. VPS52 contributes to the *spatial coordination and assembly* of the GARP complex, regulates protein and glycoprotein sorting, is important for normal Golgi physiology and glycosylation processing, and is essential for vesicle trafficking pathways required for normal cell function. Disruption of VPS52 leads to sorting defects, abnormal glycosylation, mistargeting of hydrolases, and is linked to certain developmental and possibly neurodegenerative diseases. VPS52 has no known direct drug interactions or established mechanisms of therapeutic modulation.
