Overview

Vacuolar protein sorting-associated protein 72 homolog (VPS72) is a protein encoded by the VPS72 gene in humans. It functions as a deposition-and-exchange histone chaperone specific for the incorporation of the histone variant H2A.Z (H2AFZ) into nucleosomes. VPS72 is a core component of both the SRCAP chromatin remodeling complex, which mediates exchange of H2A/H2B for H2A.Z/H2B in nucleosomes, and the TRRAP/TIP60 histone acetyltransferase complex, which is involved in transcriptional regulation, DNA repair, and apoptosis. VPS72 plays important roles in chromatin structure modulation, regulation of gene expression, DNA repair pathways, and possibly long-term hematopoietic stem cell regulation. Overexpression has been linked to increased proliferation, invasion, and migration in hepatocellular carcinoma, partially through regulation of PI3K/AKT signaling by binding to lysine acetyltransferase 5 (KAT5)[1][2][5].\n\nNo currently known therapeutic drugs are documented to directly target VPS72, nor are there established mechanisms of action or biomarker uses based on current literature.
