Overview

Vacuolar protein sorting-associated protein VTA1 homolog (VTA1) is a cofactor protein primarily involved in the endosomal sorting complex required for transport (ESCRT) pathway, particularly in regulating the activity of the AAA-ATPase Vps4, which is required for recycling ESCRT-III complexes during multivesicular body (MVB) formation[1][2][4][5]. VTA1 promotes the oligomerization and activity of Vps4 by stabilizing its ATP-dependent double-ring assembly, and interacts with other ESCRT-III accessory proteins such as Vps60 and Did2[1][3][4]. VTA1 is essential for efficient cargo sorting from endosomes and proper turnover of proteins like the epidermal growth factor receptor (EGFR)[2]. Mutations or dysregulation of the MVB/ESCRT pathway proteins, including VTA1, can broadly influence processes like receptor downregulation, virus budding, cytokinesis, and potentially neurological or lysosomal disorders, but VTA1 itself is not a primary disease target and is not known to be directly modulated by therapeutic drugs[2][4]. Its key function is to ensure accurate protein sorting and membrane remodeling within the endosomal system via regulation of molecular machines essential for vesicle trafficking[1][2][4].
