Overview

Vacuole membrane protein 1 (VMP1) is an evolutionarily conserved integral membrane protein primarily localized to the endoplasmic reticulum (ER)[1][3]. It plays a critical role in the initiation of autophagy by facilitating the formation of autophagosomes and by regulating contact sites between the ER and other organelles, including mitochondria and endosomes/endolysosomes[1][3]. VMP1 consists of six hydrophobic transmembrane regions, with both N- and C-termini exposed to the cytosol[1]. Its C-terminal domain interacts directly with Beclin 1, a key autophagy regulator, and enables the recruitment of the class III PI3K complex necessary for autophagosome formation[4]. VMP1 also controls intracellular calcium homeostasis and impacts organelle morphology and membrane trafficking. Loss of VMP1 function causes broad defects, including impaired autophagy, abnormal mitochondrial morphology, disrupted Golgi and ER integrity, and increased organelle contacts, which have been linked to pathological processes in cancer, neurodegenerative diseases, hepatitis, and pancreatitis[1][3][4][5]. Dysregulation or mutations of VMP1 are associated with protein aggregation characteristic of neurodegenerative disease and with progression of pancreatic and other cancers[3][1]. While not yet a direct drug target, VMP1 is considered a marker of autophagic flux and disease state, and a potential future therapeutic target for diseases involving defective autophagy and organelle homeostasis[3][4].
