The vaginal mucosal tissue is the internal lining of the vagina, primarily composed of a non-keratinized stratified squamous epithelium and a supporting lamina propria (StatPearls, 2023). Its primary biological functions include acting as a protective barrier against mechanical stress and pathogens, maintaining an acidic environment through glycogen metabolism by commensal bacteria, and providing lubrication (NIH, 2022). While it is an anatomical structure rather than a single molecular target, it contains various receptors, such as estrogen and progesterone receptors, which are targeted by hormonal therapies to treat conditions like vulvovaginal atrophy (PubMed, 2021). Additionally, the mucosa is a significant site for the delivery of anti-infectives and microbicides intended to prevent the transmission of HIV and other sexually transmitted infections (Journal of Controlled Release, 2019). The tissue's unique environment, including its pH and microflora, significantly influences drug stability and absorption (PubChem, 2023).
Drugs interact with the vaginal mucosal tissue through local absorption across the epithelial barrier, binding to specific receptors (e.g., estrogen receptors) within the tissue, or by altering the local environment such as pH and microbial flora (PubMed, 2021; StatPearls, 2023).
