Variable charge X-linked protein 1 (VCX1) is a member of the VCX/Y gene family, which encodes small, highly charged nuclear proteins primarily expressed in male germ cells. Located on the X chromosome at p22.31, the VCX gene cluster is characterized by significant copy number polymorphism among individuals and plays a critical role in spermatogenesis and neuronal development. VCX1 is involved in fundamental cellular processes including chromatin organization, ribosome assembly, and the regulation of mRNA stability, specifically by inhibiting mRNA decapping. Clinically, deletions in the Xp22.3 region encompassing the VCX genes are associated with X-linked ichthyosis and have been implicated in non-specific mental retardation. Due to its restricted expression in the testis and aberrant expression in various malignancies such as lung cancer, VCX1 is classified as a Cancer/Testis (CT) antigen, making it a potential candidate for targeted cancer immunotherapies and vaccine development.
