Overview

Variable charge Y-linked protein 1 (VCY) is a small, highly charged, nuclear protein encoded by a testis-specific gene family present on both the X and Y chromosomes, expressed exclusively in the nuclei of male germ cells, especially during spermatogenesis[1][3][2]. Members of this gene family share high sequence identity, differing mainly in a repetitive sequence region between X- and Y-linked copies[3][2]. VCY/VCX proteins are enriched in the nuclei and nucleoli of spermatogenic cells and are hypothesized to play a regulatory role in ribosome assembly during spermatogenesis, though their precise biological function remains unknown[1][3][2]. The VCY1 gene has been implicated in spermatogenic failure and is considered part of the cancer/testis antigen repertoire, but no drugs or established mechanisms of action as a therapeutic target are known[5].
