Overview

The variola virus is the causative agent of smallpox, a highly contagious and often deadly disease that has been eradicated globally since 1977. It is a large, brick-shaped DNA virus that replicates entirely within the cytoplasm. Transmission occurs via respiratory droplets or contaminated materials. While eradicated, it remains a potential threat as a biological weapon.

Mechanism of action

Vaccinia virus vaccine provides immunity. Cidofovir and brincidofovir are antiviral agents that inhibit viral DNA polymerase. Tecovirimat inhibits the VP37 protein, preventing the formation of extracellular virus.
