Variola virus protein A33 is a 23-28 kDa type II membrane glycoprotein essential for the life cycle of the virus that causes smallpox [1, 8]. It is specifically located on the outer membrane of the Extracellular Enveloped Virus (EEV), a form of the virus responsible for long-range dissemination within the host [1, 16]. A33 plays a critical role in viral pathogenesis by facilitating the formation of actin tails and the efficient spread of the virus from cell to cell [1, 17]. It also mediates the proper trafficking of other essential envelope proteins, such as B5, to the EEV membrane [1, 8]. Structurally, A33 contains a C-type lectin-like domain (CTLD) and exists as a disulfide-bonded homodimer on the viral surface [8, 17]. Because it is a major target for protective antibody responses, A33 is a primary candidate for next-generation subunit and DNA vaccines, such as the "4pox" vaccine, which aim to provide safer alternatives to traditional live-virus vaccines [2, 16]. While no drugs currently target A33, experimental monoclonal antibodies like A27D7 have shown potent neutralizing activity against multiple orthopoxviruses by blocking EEV-mediated spread [6, 16]. The protein is highly conserved among orthopoxviruses, although minor sequence variations can affect the cross-reactivity of certain therapeutic antibodies [2]. Targeting A33 in combination with other viral proteins is considered a robust strategy for developing broad-spectrum orthopoxvirus countermeasures [16].
