Vascular endothelial cell differentiation is a fundamental biological process through which precursor cells, such as angioblasts or stem cells, commit to an endothelial lineage and mature into functional cells lining the blood vessels (Gene Ontology, GO:0035864). This process is orchestrated by a complex network of signaling pathways, most notably the Vascular Endothelial Growth Factor (VEGF) and Notch pathways, which regulate cell fate, proliferation, and organization into tubular structures (PubMed, PMID: 21606367). In a therapeutic context, this process is not a single molecular target but rather a physiological outcome modulated in various diseases. Pathological upregulation of endothelial differentiation and angiogenesis is a hallmark of cancer and age-related macular degeneration, leading to the development of clinical inhibitors like bevacizumab and ranibizumab (NIH, National Cancer Institute). Conversely, promoting this process is a primary goal in regenerative medicine and the treatment of ischemic cardiovascular diseases to restore blood flow to damaged tissues (PubMed, PMID: 25173165).
