Overview

The vascular endothelial cell membrane is the specialized lipid bilayer that forms the luminal surface of all blood vessels, acting as a dynamic barrier between the blood and the vessel wall (Molecular Biology of the Cell, 4th ed.). It is not a single molecular target but a complex cellular structure that integrates mechanical and chemical signals to maintain vascular homeostasis (Arteriosclerosis, Thrombosis, and Vascular Biology, 2009). The membrane contains a vast array of proteins, including receptors for growth factors (e.g., VEGFR), adhesion molecules (e.g., ICAM-1), and ion channels that regulate vascular tone and permeability (Circulation, 2005). Dysfunction of this membrane is a central feature in the development of atherosclerosis, hypertension, and diabetic complications (Nature Reviews Cardiology, 2021). While many drugs target specific proteins located on or within this membrane, the membrane itself is too broad and heterogeneous to be classified as a discrete therapeutic target (Pharmacological Reviews, 2017). Consequently, it is considered a cellular component rather than a specific molecular entity for drug discovery purposes.
