The vascular endothelial growth factor A (VEGF-A) signaling pathway is a critical regulator of blood vessel formation and function. VEGF-A primarily acts through its receptors on endothelial cells to mediate vasculogenesis and angiogenesis. This pathway is essential for normal vascular development, tissue repair, and also plays significant roles in pathological conditions such as cancer, age-related macular degeneration, rheumatoid arthritis, and neurodegenerative diseases. The pathway regulates endothelial cell proliferation, migration, survival, and vascular permeability. Aberrant activation or regulation leads to tumor neovascularization, ocular diseases, rheumatoid arthritis, and neurodegeneration. Therapeutic targeting focuses on blocking either the ligand itself or its interaction with key receptors.
