Vascular Endothelial Growth Factor C (VEGF-C) is a secreted growth factor that promotes lymphangiogenesis and angiogenesis. It primarily signals through VEGFR-3 on lymphatic endothelial cells (LECs), stimulating their proliferation, migration, and survival. VEGF-C also binds to VEGFR-2 on blood vessel endothelial cells, contributing to angiogenesis and increased vascular permeability. Proteolytic processing of the VEGF-C propeptide is essential for its full activity and interaction with Neuropilin-2. VEGF-C is implicated in various physiological and pathological processes, including embryonic development, tissue repair, lymphedema, and cancer metastasis, making it a potential therapeutic target.
VEGF-C binds to VEGFR-3 (primary) and VEGFR-2 (secondary) receptors, initiating downstream signaling cascades that promote lymphatic and blood vessel growth, survival, and permeability. Binding to Neuropilin-2 enhances VEGFR-3 signaling.
