Overview

Vascular tone refers to the degree of constriction or dilation in blood vessels, determined primarily by the contractile state of vascular smooth muscle cells within the vessel wall[1][7][9]. It is a physiological property rather than a discrete molecular entity. Vascular tone plays an essential role in regulating systemic and local blood pressure, ensuring adequate tissue perfusion under varying physiological conditions such as exercise or stress[1]. Its regulation involves complex interactions between neural inputs (sympathetic/parasympathetic nervous systems), hormonal signals (adrenaline, angiotensin II), local metabolic factors, and endothelial-derived mediators like nitric oxide[1][4][8]. Abnormalities in vascular tone contribute to diseases such as hypertension when excessive vasoconstriction occurs or shock when there is excessive vasodilation[5]. Many drugs—including alpha agonists/antagonists, calcium channel blockers, ACE inhibitors, nitrates—modulate vascular tone indirectly by acting on receptors or enzymes involved in these regulatory pathways. However, "vascular tone" itself is not a single therapeutic target but rather an emergent property resulting from multiple underlying molecular targets. Because "vascular tone" does not refer to any specific molecule/receptor/enzyme/transporter, it should not be considered a canonical drug target. Instead, it describes the net effect produced by various cellular mechanisms within the vasculature. “Vascular tone refers to the degree of constriction experienced by a blood vessel relative to its maximally dilated state… primarily influenced by smooth muscle cells in the vessel walls.”[9][1] “The regulation of vascular smooth muscle contraction… alters blood vessel volume and local blood pressure… Excessive vasoconstriction causes hypertension while excessive vasodilation leads to hypotension.”[5] In summary: "Vascular tone" is not itself a valid molecular drug target but rather describes an important physiological parameter controlled by numerous underlying targets.
