The vasopressin receptor 1B (V1BR), also known as vasopressin 3 receptor (VPR3) or antidiuretic hormone receptor 1B, is a G protein-coupled receptor that primarily acts as a cell surface receptor for arginine vasopressin. It is highly expressed in corticotroph cells of the anterior pituitary gland, where it plays a key role in stimulating adrenocorticotropic hormone (ACTH) release. It is also found in various brain regions, pancreas, and adrenal glands, influencing insulin secretion and catecholamine release. Centrally expressed AVPR1b modulates social behaviors such as aggression and social recognition. Genetic variations within AVPR1B have been linked to susceptibility to depression. Selective antagonists for V1BR have demonstrated anti-aggressive effects in animal models and exhibit antidepressant-like or anxiolytic properties.
Selective antagonists for V1BR block the binding of arginine vasopressin, preventing activation of phospholipase C and subsequent increase in intracellular calcium levels.
