Vault RNA 1-1 (VTRNA1-1) is a small non-coding RNA that forms part of the vault particle, a large cytoplasmic ribonucleoprotein complex. VTRNA1-1 is best characterized for its regulatory role in modulating apoptosis; it inhibits both intrinsic and extrinsic apoptotic pathways, facilitating cell survival under conditions of stress such as nutrient deprivation. VTRNA1-1 exerts its function by modulating major pro-survival signaling pathways, including PI3K/Akt and ERK1/2 MAPK, and by acting as a riboregulator through direct binding to the autophagic receptor p62/SQSTM1, thereby affecting autophagy and protein homeostasis. Unlike many well-known therapeutic targets, VTRNA1-1 is not a receptor, enzyme, or transporter, but a non-coding RNA with important roles in cellular stress responses and cancer biology[1][2][3][4][5].
