Overview

VCAN antisense RNA 1 (VCAN-AS1) is a long non-coding RNA (lncRNA) located on chromosome 5 (chr5:83531352-83581320, hg38), functioning as a non-protein-coding transcript[2]. Recent research implicates VCAN-AS1 in cancer, especially gastric cancer. It is significantly upregulated in gastric cancer tissues and cell lines, with higher expression levels associated with poor prognosis. Mechanistically, VCAN-AS1 promotes cancer progression by interacting with the RNA-binding protein HuR (ELAVL1), which increases the stability of F11 receptor (F11R) mRNA, leading to enhanced cell proliferation and migration[1][3]. Knockdown of VCAN-AS1 suppresses tumor growth in vitro and in vivo. In addition to gastric cancer, VCAN-AS1 has been reported to promote cancer through other pathways, such as modulating the miR-106a-5p/STAT3 pathway in breast cancer and participating in ceRNA networks involved in diabetic kidney disease[1][3]. There are no approved drugs directly targeting VCAN-AS1, but it is considered a potential therapeutic target and a prognostic biomarker, particularly in gastric cancer[1][3].
