VCP nuclear cofactor family member 2 (VCF2) is a member of the FAM104 protein family and functions as a nuclear cofactor for the ATPase p97 (also known as VCP or Cdc48). VCF2 binds to p97 directly through a non-canonical, alpha-helical motif and is required for the efficient nuclear import of p97. Loss of VCF2 (and its family member VCF1) causes reduced nuclear levels of p97, slow cell growth, and increased cell sensitivity to p97 inhibitors and DNA damage, indicating that VCF2 is a critical regulator of nuclear p97 function. VCF2 forms complexes with p97 cofactors such as UFD1, NPL4, FAF1, and UBXN2B, linking it to diverse nuclear quality control and protein degradation processes, including DNA replication and repair. There is no evidence it directly functions as a receptor, enzyme, transporter, or classic drug target[1][2][4][6].
