Overview

The VEGFA gene promoter is a DNA regulatory region upstream of the VEGFA coding sequence, responsible for directing transcription of the VEGFA gene. The promoter lacks a canonical TATA box but contains several conserved binding sites for transcription factors, including AP1, AP2, Sp1, and HIFs (Hypoxia Inducible Factors), which mediate responses to stimuli such as growth factors, cytokines, hormones, tumor suppressors, and most importantly, hypoxia[3][6]. Hypoxic environments, as seen in cancer or ischemic tissue, induce HIF-1 and HIF-2 binding to the promoter’s hypoxia response elements, leading to upregulation of VEGFA transcription[3][6]. Alternative promoter usage and interplay with factors like WT1 also modulate VEGFA expression in different contexts[3][6]. The VEGFA promoter is not itself a druggable entity in standard therapeutics, although it is of interest in experimental gene therapy and regulatory biology.
