Overview

The Venezuelan equine encephalitis virus (VEEV) envelope polyprotein is a critical structural component of the VEEV virion, an alphavirus responsible for severe neurological disease in humans and equines. This polyprotein is post-translationally cleaved by host and viral proteases into four distinct subunits: E3, E2, 6K, and E1. The E2 glycoprotein is primarily responsible for host cell receptor binding, while the E1 glycoprotein functions as a class II fusion protein that mediates the merger of the viral envelope with the host endosomal membrane under acidic conditions. Due to its essential role in the viral life cycle, the envelope polyprotein is the primary target for neutralizing antibodies and vaccine development. Therapeutic strategies typically focus on blocking E2-mediated attachment or preventing the conformational changes in E1 required for membrane fusion. Ongoing research aims to develop potent monoclonal antibodies and small-molecule inhibitors to mitigate the high morbidity and potential mortality associated with VEEV outbreaks and its potential use as a biological threat agent.
