Venous smooth muscle alpha-adrenergic receptors are G protein-coupled receptors (GPCRs) located on the smooth muscle cells of veins. These receptors mediate the contractile response of venous smooth muscle to catecholamines, primarily norepinephrine and epinephrine, released by sympathetic nerve terminals or circulating in the blood. Activation of these receptors causes contraction of venous smooth muscle—vasoconstriction—which reduces vein diameter, increases venous return to the heart, and helps maintain blood pressure during stress conditions. There are two principal types of alpha-adrenergic receptors relevant to venous smooth muscle: Alpha-1 (α1-AR), which are classic postsynaptic receptors leading to vasoconstriction, and Alpha-2 (α2-AR), present both pre- and post-junctionally, also mediating vasoconstriction, though less prominently than α1.
