Overview

VENT homeobox pseudogene 4 (VENTXP4) is a member of the Ventx family of homeobox gene-derived pseudogenes in the human genome[3][4]. VENTXP4, along with several other VENTX pseudogenes (VENTXP1-7), arose via retrotransposition from the functional VENTX gene (also previously known as VENTX2), and does not encode a functional protein due to accumulated disabling mutations[3]. Unlike protein-coding homeobox genes or certain functional retrogenes, there is no published evidence that VENTXP4 is transcribed or contributes to biological pathways, development, or disease in humans[3]. As a processed pseudogene, it lacks a promoter and the necessary regulatory elements for functional expression[3]. Therefore, VENTXP4 is not considered a therapeutic target, biomarker, or gene with established biological relevance. The functional gene of the family, VENTX, encodes a transcriptional regulator involved in human hematopoiesis and leukemogenesis, but the pseudogenes, including VENTXP4, are non-functional[1][3]. The nomenclature VENTXP4 replaces prior symbols such as VENTX2P4 to reflect its origin as a pseudogene derived from VENTX[3][4]. No clinical, pharmacological, or diagnostic relevance for VENTXP4 is currently known[3][4]. VENTXP4 should not be confused with VENTX (the protein-coding gene and functional homeobox transcription factor), which is implicated in leukemia and hematopoietic differentiation[1][2][3].
