Ventricular heart development-associated long noncoding RNA (VHRT, also called VENTHEART, LINC01405, MASCC1, among other names), is a primate-conserved, ventricular myocyte-specific long noncoding RNA expressed in fetal and adult human ventricles but not in atria. Functional studies using human pluripotent stem cell-derived cardiomyocytes showed that VHRT is necessary for the specification and maturation of ventricular cardiomyocytes, regulating genes involved in cardiac muscle structure and function. CRISPR-mediated knockout of VHRT disrupts sarcomere formation and alters the ventricular cardiomyocyte gene expression program without affecting atrial cardiomyocytes, and leads to a prolonged ventricular action potential duration—demonstrating a unique and essential role in ventricular, but not atrial, cardiac development. VHRT does not correspond to a typical therapeutic target class such as a receptor, enzyme, or transporter, and has no known drug interactions or direct disease biomarker applications as of current literature[1][2].
