**Vesicle-associated membrane protein 4 (VAMP4)** is a member of the VAMP/synaptobrevin family, classified under the broader group of SNARE (Soluble NSF Attachment Protein Receptor) proteins, which are crucial mediators of membrane fusion events. VAMP4 is primarily localized to the trans-Golgi network (TGN), endosomes, and clathrin-coated vesicles. Its major role is facilitating *trans*-Golgi network-to-endosome transport, and it forms SNARE complexes with proteins such as syntaxin 6, STX16, and AP1M1[1][3][4][5]. In neuronal cells, VAMP4 is essential for activity-dependent bulk endocytosis (ADBE)—a process important for maintaining synaptic vesicle pools during periods of intense synaptic activity[2]. VAMP4 is not a known classical drug target, nor is it associated with any specific therapeutic agent or clinical biomarker use currently. Its dysfunction may contribute to cellular trafficking defects, but no specific human disease associations (such as cancer or neurodegeneration) are well established at present[4].
