Overview

Vesicular monoamine transporter type 1 (VMAT1) is an integral membrane protein responsible for transporting monoamines—such as norepinephrine, epinephrine, dopamine, serotonin, and histamine—from the cytosol into synaptic or secretory vesicles within neuroendocrine cells. This process is essential for the storage and regulated release of these neurotransmitters. VMAT1 operates via secondary active transport as an antiporter, utilizing the proton gradient generated by vesicular H+-ATPase to drive monoamine uptake against a steep concentration gradient. It is predominantly expressed in large dense-core vesicles of neuroendocrine cells such as chromaffin cells in adrenal medulla and enterochromaffin cells in the gut. Certain genetic polymorphisms affect transporter efficiency and have been associated with altered emotional processing circuits in humans.
