Overview

The Vibrio cholerae O1 Ogawa O-antigen is a critical surface polysaccharide component of the lipopolysaccharide (LPS) found in the Ogawa serotype of Vibrio cholerae O1, the primary causative agent of epidemic cholera (Chatterjee & Chaudhuri, 2003; https://doi.org/10.1016/S0925-4439(03)00061-1). Structurally, it consists of a homopolymer of alpha(1-2)-linked 4-amino-4-deoxy-D-glycero-D-talo-octonic acid, distinguished from the Inaba serotype by the presence of a 2-O-methyl group on the non-reducing terminal sugar (Villeneuve et al., 2000; https://doi.org/10.1073/pnas.97.15.8433). This antigen plays a vital role in bacterial pathogenesis and is the dominant target for protective vibriocidal antibodies in the human host (Harris et al., 2012; https://doi.org/10.1016/S0140-6736(12)60436-X). Therapeutic strategies primarily involve oral cholera vaccines (OCVs) that utilize inactivated whole cells or purified O-antigen conjugates to elicit mucosal immunity (WHO, 2017; https://www.who.int/publications/i/item/who-wer9234). Monitoring immune responses to this specific antigen is essential for evaluating vaccine efficacy and understanding the epidemiology of cholera outbreaks.
