Vibrio cholerae O1 surface O-antigen lipopolysaccharide (LPS) is a major structural and immunogenic component of the outer membrane of the Gram-negative bacterium Vibrio cholerae, the causative agent of cholera (Chatterjee & Chaudhuri, 2011, PubMed). The molecule is composed of three regions: Lipid A, a core oligosaccharide, and the O-specific polysaccharide (OSP), which is a homopolymer of 4-amino-4-deoxy-L-glycero-D-manno-heptopyranose (PubChem CID 11966243). The OSP defines the O1 serogroup and is further classified into Ogawa and Inaba serotypes based on the presence of a methyl group. As the primary target of the protective immune response, O1 LPS is the central antigen in both killed whole-cell and live-attenuated oral cholera vaccines, such as Dukoral and Vaxchora (Wierzba et al., 2015, PubMed). Antibodies directed against the OSP, particularly vibriocidal antibodies, are the best-characterized correlates of protection against cholera infection (Harris et al., 2012, PubMed). Additionally, O1 LPS serves as a critical diagnostic marker for identifying and serotyping V. cholerae strains in clinical and environmental samples (WHO, 2017).
