Overview

Villin-1 is a calcium-regulated actin-binding protein encoded by the VIL1 gene in humans[1][3][4][6]. It is a dominant component of the brush border cytoskeleton, especially in epithelial cells of the gastrointestinal tract and other organs with microvilli[1][3][4][6]. Villin-1 is specialized in modulating actin dynamics through capping, severing, nucleating, and bundling actin filaments, impacting the structure and morphogenesis of microvilli[2][4]. Its function is essential for maintaining intestinal epithelial cell integrity, supporting cell migration, wound repair, and regulating apoptosis, notably protecting against gastrointestinal injury[3][5]. Its highly regulated expression and location make it a sensitive marker for epithelial cells and diseases affecting the intestine[3][5][6]. Villin-1 is not considered a classical drug target (such as a receptor or enzyme), and no drugs are known to modulate it directly, but its dysregulation is linked to various gastrointestinal disorders[5].
